碧云天生产的抗荧光淬灭封片液(含DAPI) (Antifade Mounting Medium with DAPI)是一种含有细胞核蓝色荧光染料DAPI并能减缓荧光淬灭的封片试剂。本封片液不仅可以减缓各种常见荧光染料的荧光淬灭,还能在封片的同时完成对细胞核的DAPI染色。
本封片液内含DAPI,DAPI的英文全称为2-(4-Amidinophenyl)-6-indolecarbamidine dihydrochloride,中文名称为4',6-二脒基-2-苯基吲哚,也称DAPI dihydrochloride,分子式为C16H15N5·2HCl,分子量为350.25,CAS Number 28718-90-3。
DAPI的最大激发光波长为340nm,最大发射光波长为488nm;DAPI和双链DNA结合后,最大激发光波长为364nm,最大发射光波长为454nm。DAPI和双链DNA结合后的激发光谱和发射光谱参考图1。

DAPI是一种可以穿透细胞膜的蓝色荧光染料,和双链DNA结合后可以产生比DAPI自身强20多倍的荧光,和EB(ethidium bromide)相比,对双链DNA的染色灵敏度要高很多倍。
DAPI染色常用于细胞凋亡检测,染色后用荧光显微镜观察或流式细胞仪检测。DAPI也常用于普通的细胞核染色以及某些特定情况下的双链DNA染色。
本产品用于细胞染色的效果图参考图2。

本品已加DAPI,可同时完成细胞核染色和封片,并能减缓荧光淬灭,实验操作更简单,只需在封片时用移液器滴一滴本产品在样品上,盖上盖玻片即可完成操作。

按照每个样品封片需要50微升计算,5ml本产品足够用于100个样品的细胞核染色和封片,25ml本产品可以用于500个样品的细胞核染色和封片。

注意事项:
需自备盖玻片与载玻片,也可向碧云天订购盖玻片(FCGF18)(FCGF20)(FCGF22)(FCGF24)(FCGF32)(FCGF40)(FCGF50)(FCGF60)与载玻片(FSL004)(FSL005)(FSL006)(FSL011)(FSL012)(FSL051)。
荧光物质均易发生淬灭,染色后的样品宜避光保存。可以向碧云天选购各种型号的载玻片存储盒(FBX001)(FBX002)(FBX003)(FBX005)(FBX011)(FBX012)(FBX024)(FBX025)(FBX049)(FBX050)(FBX099)(FBX100)。
本封片液中DAPI浓度经过碧云天的优化,确保可以满足各种常规染色的需要。如需单独使用特定浓度的DAPI,请选购碧云天的DAPI(C1002)。
在使用抗淬灭封片液的情况下可以减缓荧光淬灭,但仍宜尽量避光。
DAPI对人体有刺激性,操作时请小心,并注意适当防护以避免直接接触人体或吸入体内。

使用说明:
1. 贴壁细胞样品:
a. 染色完毕后,吸尽液体。
b. 滴一滴抗荧光淬灭封片液于载玻片上,盖上贴有细胞的盖玻片,让细胞接触封片液,尽量避免气泡。
c. 随后即可在荧光显微镜下观察细胞样品。
2. 组织切片:
a. 染色完毕后,吸尽液体。
b. 滴一滴抗荧光淬灭封片液于组织切片上,盖上盖玻片,让切片接触封片液,尽量避免气泡。
c. 随后即可在荧光显微镜下观察组织切片。
3. 其它样品:
其它样品参考细胞样品或组织切片进行操作。
