AVP Knockout HEK293T Trizol Lysate (AVP基因敲除HEK293T细胞Trizol裂解液)是通过RNP方法获得的敲除多克隆HEK293T细胞的Trizol裂解液。该细胞中目的基因的敲除效率已经通过PCR扩增测序验证。本产品可用于该目的基因敲除后其信号通路相关RNA表达的研究。
由于本细胞是通过RNP技术获得的多克隆细胞池(Cell pool)，该方法无需病毒包装以及基因组DNA整合，免疫原性小，细胞不携带荧光或者嘌呤抗性，敲除效率可达到70%，已经足以进行很多目的基因的生物学功能研究了。如果希望获得100%基因敲除的细胞，可以自行使用本产品筛选单克隆细胞，通过稀释法将细胞稀释至单细胞水平，培养形成单克隆群，然后提取DNA进行测序验证其单一性和序列正确性，或者委托碧云天进行单克隆细胞株的筛选服务。
本Trizol裂解液用于实验时，建议同时选购293T (人胚肾细胞) (C6008)用于对照细胞Trizol裂解液的制备。

| Gene Summary | This gene encodes a member of the vasopressin/oxytocin family and preproprotein that is proteolytically processed to generate multiple protein products. These products include the neuropeptide hormone arginine vasopressin, and two other peptides, neurophysin 2 and copeptin. Arginine vasopressin is a posterior pituitary hormone that is synthesized in the supraoptic nucleus and paraventricular nucleus of the hypothalamus. Along with its carrier protein, neurophysin 2, it is packaged into neurosecretory vesicles and transported axonally to the nerve endings in the neurohypophysis where it is either stored or secreted into the bloodstream. The precursor is thought to be activated while it is being transported along the axon to the posterior pituitary. Arginine vasopressin acts as a growth factor by enhancing pH regulation through acid-base transport systems. It has a direct antidiuretic action on the kidney, and also causes vasoconstriction of the peripheral vessels. This hormone can contract smooth muscle during parturition and lactation. It is also involved in cognition, tolerance, adaptation and complex sexual and maternal behaviour, as well as in the regulation of water excretion and cardiovascular functions. Mutations in this gene cause autosomal dominant neurohypophyseal diabetes insipidus (ADNDI). This gene is present in a gene cluster with the related gene oxytocin on chromosome 20. |
