Background
|
The IL-17 family of cytokines consists of IL-17A-F, and their receptors include IL-17RA-RE. IL-17 cytokines are produced by a variety of cell types including the Th17 subset of CD4+ T cells, as well as subsets of γδ T cells, NK cells, and NKT cells. IL-17A and IL-17F, the most well-studied of the IL-17 cytokines, contribute to fungal and bacterial immunity by inducing expression of proinflammatory cytokines, chemokines, and antimicrobial peptides. In addition, IL-17A contributes to the pathogenesis of several autoimmune diseases. IL-17E promotes Th2 cell responses. The roles of IL-17B, IL-17C, and IL-17D are less clear, however these family members also appear to have the capacity to induce proinflammatory cytokines. IL-17 receptors have an extracellular domain, a transmembrane domain, and a SEFIR domain. They are believed to signal as homodimers, heterodimers, or multimers through their SEFIR domain by recruiting the SEFIR domain-containing adaptor Act1. Unlike most cytokines that signal through Jak/STAT pathways, IL-17 signaling results in NF-κB activation.
IL-17F is a cysteine-linked proinflammatory cytokine that can dimerize with itself or form heterodimers with the IL-17 family member that it shares 50% homology with, IL-17A. Although mainly produced by Th17 cells, IL-17F expression has been observed in several cell types including activated CD8+ T cells, γδ T cells, NKT cells, B cells and LTi cells. IL-17F binds to a heterodimeric receptor consisting of IL-17RA and IL-17RC, which upon binding induces the TRAF6-mediated activation of TAK and the Erk1/2 MAP kinase pathway. This induces the expression of numerous inflammatory chemokines and cytokines including IL-1β, IL-6, IL-8, and MIP-1β along with increased adhesion molecule expression in human airway epithelial cells, vein endothelial cells, and fibroblasts. IL-17F has been linked with asthma and other autoimmune diseases including rheumatoid arthritis, multiple sclerosis, psoriasis, and inflammatory bowel disease.

保存条件：
IL17F Rabbit Polyclonal Antibody -20ºC保存，Western一抗稀释液-20ºC或4ºC保存，一年有效。Western一抗稀释液优先推荐4ºC保存，长期不使用可以考虑-20ºC保存，但冻融可能会导致出现轻微的浑浊和少量不溶物。
注意事项：
如果本抗体用于Western blot (WB)、免疫荧光(IF)、免疫细胞化学(ICC)等实验，请注意回收使用过的稀释抗体。回收的抗体通常至少可以重复使用5-10次。稀释后的抗体，包括已经使用过的稀释抗体，请4℃保存。
回收后重复使用的抗体，使用方法同新鲜稀释的抗体。如果在重复使用过程中发现抗体出现轻微混浊现象，可以10,000g离心1-3分钟，取上清用于后续检测。如果回收的抗体出现明显的絮状物或长霉长菌等情况，则可以考虑废弃该抗体。
提供的Western一抗稀释液也可以用于免疫荧光(IF)、免疫组化(IHC)、免疫细胞化学(ICC)等适当用途。如果希望获得最佳的检测效果，请考虑使用上述检测专用的一抗稀释液。
