Gaussia luciferase is a novel luciferase derived from a large marine copepod (Gaussia princeps) from Hawaiian waters. It is a monomeric enzyme with a small molecular weight of 20kDa and a secretory signal peptide, and can be secreted outside the cell through the endoplasmic reticulum.

Advantages:
▶   As a natural secreted luciferase, it can be detected by analyzing the supernatant directly without the need of cell lysis, making the assay convenient, rapid, and more suitable for high-throughput assay of reporter genes at different time points.
▶   This assay method generates chemiluminescence with intensity which is more than 1000 times of the luminescent signal generated by firefly luciferase and sea kidney luciferase assays, providing higher detection sensitivity.
▶   This assay method does not require ATP and is not affected by ATP either.
▶   The assay reaction is stable, highly tolerant of temperature and pH changes, and compatible with a wide variety of culture media.

Gaussia-Lumi™ Gaussia Luciferase Reporter Gene Assay Kit (RG072)

Both of these kits are designed for the assay of secreted, non-ATP-dependent Gaussia luciferase (Gluc) with coelenterazine as the substrate. RG072 can be added directly into the cell culture at an equal volume, without the need of cell collection or centrifugation to obtain the supernatant. RG021 is suitable for detecting the luciferase activity of cell culture supernatant, cell lysate or purified luciferase.

Product Features:
High luminescence intensity: For the same cell supernatant sample, the luminescence generated by using the RG021 is about 20% stronger than that of Competitor T (Figure 1A); for the same cell lysate sample, the luminescence from RG021 is 10-50 times stronger than that of Competitor T.
Stable luminescence signal: The luminescent signal generated by using the RG021 fluctuates no more than 15% in 10 minutes, no more than 30% in 30 minutes, with a half-life time of more than 60 minutes (Figure 1C). The luminescent signal generated by using the RG072 is basically constant in 30 minutes, with no more than 10% fluctuation in 60 minutes and no more than 20% fluctuation in 120 minutes (Figure 2B), with a half-life time of about 5 hours. RG072 especially suitable for high-throughput assays of Gaussian luciferase activity in 96- or 384-well plates.
Rapid assay: When using RG021, mix 5-20µl of cell supernatant and the assay working solution, incubate for 5-10 minutes, and measure the luminescence. While using RG072, the cell cultured can be directly mixed with the assay working solution, and the luminescence can be measured directly after 5-10 minutes of incubation. Both kits provide the simple and rapid assay of Gaussia luciferase activity.
Good stability: The stability of the assay buffer is very good. Repeated freeze-thaws up to 10 cycles and storage at 4℃ for 7 days or 37℃ for 1 day have basically no effect on the assay result. The performance of assay buffer stored at 37℃ for 3 days decreases by no more than 5%, while the performance of the assay substrate stored at 4ºC for 1 week and at room temperature for 1 day decreases by no more than 10%. The assay subrstrate can still retain more than 80% of original performance after storage at room temperature for 3 days and 37ºC for 1 day.
Flexible to use: These kits are not only suitable for analyzing a small number of samples, but also suitable for thigh-throughput assays. RG021 can be used to analyze both cell supernatant and cell lysate. RG072 is suitable for analyzing whole cell culture, cell cuture supernatant, and cell lysate as well.
Good compatibility: These kits are compatible with a variety of common culture media. Phenol red, fetal calf serum or calf serum within 10%, DMSO or ethanol within 2%, and luciferase inhibitors in normal culture media have no obvious effect on the assay result, nor do commonly used salts or metal ions at normal concentrations.
