Beyotime has previously launched several one-step luciferase assay systems, such as Bio-Lumi™ Firefly Luciferase Assay System for ADCC Reporter assays, Bac-Lumi™ Bacterial Firefly Luciferase Assay System for bacterial samples, and Bright-Lumi™ Firefly Luciferase Assay System, etc. These one-step assay systems are relatively more suitable for high-throughput assay of samples in 96-well or 384-well plates. To analyze samples in culture dishes, 6-well plates or other containers with more cell samples, we newly developed the two-step luciferase assay system as well as the lysis buffers that will be used conjunctly.

Enhanced Firefly Luciferase Assay System II(RG010)
These three new products are similar to the well-received Firefly Luciferase Assay System (RG005/RG006) in employing the luciferin substrate to detect the firefly luciferase activity with high luminescence signal stability.
RG009 and RG010 assay systems generate luminescence with high intensity, and are suitable for analyzing samples with low expression of firefly luciferase.
RG007 and RG005/RG006 assay systems generate luminescence with better luminescent signal stability, and are suitable for experiments requiring high signal stability.
The Assay System II products (RG007 and RG010) are lyophilized version of the corresponding products, which are extremely stable at -20℃ before reconstitution.

Product Features:
High luminescence intensity: When analyzing the same cell samples, the luminescence intensity derived from RG009/RG010 assay systems is comparable or slightly better than that from similar products, especially when analyzing samples with low expression of firefly luciferase. RG005/RG006/RG007 assays generate luminescence with intensity which is 40% of that from RG009/RG010, and can be generally used for routine assays.
Good signal stability: When analyzing the same cell samples, the stability of luminescence generated from RG005/RG006/RG007 assay system is significantly better than that from enhanced firefly luciferase assay system and similar products, with a half-life of about 40 minutes. RG005/RG006/RG007 assay systems are especially suitable for multi-well assay of firefly luciferase in a large number of samples.
Rapid assay: This series of products are easy to operate, with stable readings, and require about 20 minutes to complete the assay.
Good reagent stability: Repeated freeze-thaw of assay reagent up to 5 cycles has no significant effect on the assay result. The performance decreases by no more than 10% after 10 freeze-thaws. Even with only about 50% of original performance, these products can still be used for routine assays.

Dual Luciferase Assay System II (RG029)
To improve the accuracy of the assay, Renilla luciferase is often used as an internal reference for transfection efficiency to eliminate differences in cell numbers and transfection efficiency, and there is a need for a kit that can accurately detect both firefly and renilla luciferases simultaneously.
RG029 is similar to RG027/RG028 products in employing luciferin substrate to detect firefly luciferase activity and coelenterazine substrate to detect renilla luciferase activity, with same performances. RG027/RG028 is ready-to-use liquid, while RG029 is the lyophilized version of RG027/RG028 and extremely stable at -20℃ before reconstitution.
Product Features:
High luminescence intensity: When analyzing the same samples, the luminescence generated by the firefly lucifease reaction is 30-45% higher than that of a widely-received similar product (Competitor P) and that generated by the renilla luciferase is 20-30% higher than that of Competitor P.
Rapid assay: This product is easy to operate, with stable readings. It takes only 25 minutes in total to complete sample preparation and luciferase assay. The produced luminescence is relatively stable, with a half-life of 15 minutes for firefly luciferase assay and about 200 seconds for renilla luciferase assay.
Good assay performance: The renilla luciferase assay buffer and renilla luciferase assay working solution in this product contain substances that can inhibit more than 99.9% of the luminescence signal catalyzed by firefly luciferase, greatly improving the accuracy of the subsequent renilla luciferase activity assay.
Good stability: Firefly luciferase assay reagent II is basically unaffected by repeated freeze-thaws up to 5 cycles, and its performance decreases no more than 10% after 10 freeze-thaws. Repeated freeze-thaws of renilla luciferase assay buffer up to 10 cycles and storage at 4℃ or room temperature for 3 days have basically no effect on the assay result. Renilla luciferase assay buffer stored at 37℃ for 1 day has no more than 5% fluctuation of the assay result. Renilla luciferase assay substrate (100X) stored at 4℃ for 1 week and at room temperature for 1 day has less than 10% decrease in performance and can still retain more than 80% activity when stored at room temperature for 3 days and 37℃ for 1 day.

Besides renilla luciferase reporter, β-galactosidase reporter can also be utilized as an internal reference. In such a case, Beyotime's β-galactosidase Assay System (RG0036) should be used for the assay.
Cell Lysis Buffer for Enhanced Firefly Luciferase Assay (RG127)
Cell Lysis Buffer for Dual Luciferase Assay (RG132)
This time, we launch two new cell lysis buffers. RG127 is for firefly luciferase assays and RG132 is for dual luciferase assays.
▶ Cell lysates can be immediately assayed for luciferase activity or can be frozen for future analysis. The frozen cell lysates need to be thawed completely and equilibrated to room temperature before use.
▶ Cell lysates prepared with these two lysis buffers can be used for SDS-PAGE and Western blot analysis after protein concentration determination with the BCA protein assay method.
